Isolation of Angiotensin 1 Converting Enzyme Inhibitory Peptide from Soybean Hydrolysate

Abstract
The soybean hydrolysates prepared with various proteases were tested for the inhibitory effects against angiotensin converting enzyme (ACE). Hydrolysate treated with protease isolated from Bacillus subtilis was shown to have the highest inhibitory effect (91.6%). Degree of hydrolysis (DH) increased with increasing hydrolysis time. The maximum inhibitory effect (92.1%) was observed after hydrolysis for 5 hr. After the isolation of ACE inhibitor from the hydrolysate treated with protease from B. subtilis (IC_(50) value of 196.4 §¶/mL) through acetone fractionation, we obtained the ACE inhibitor with IC_50 value of 26.5 §¶/mL with several successive column chromatographies, Sephadex G-25, Sephadex LH-20, ODS, and Supercosil PLC-18. The inhibitor showed the competitive inhibition pattern on ACE and had a plenty of Pro, Gly, Thr, A1a, Val, Phe, and Lys.
